Nonuniform Association of Genetic Risk Scores for Intraocular Pressure

Is the nonuniform association of a genetic risk score for intraocular pressure (IOP) across quantiles of IOP attributable to a distinct type of genetic interaction?In this genetic association study including a sample of 98 235 UK Biobank participants, 14 of 98 genome-wide association study variants associated with IOP had evidence of a genotype × age interaction. There was minimal evidence for other types of interaction; however, accuracy of a genetic risk score for IOP was not improved by accounting for interactions with age.Findings suggest that genetic risk scores for IOP are more accurate in older patients, and integrating information about genotype × age interactions during construction did not improve accuracy.
