Mammalian Target Rapamycin Inhibition as a Therapeutic Target for Prevention of Proliferative Vitreoretinopathy

What were the outcomes of patients who underwent retinal detachment (RD) repair while concomitantly using systemic mammalian target rapamycin (mTOR) inhibitor (MTI) therapy?In this cohort study, 681 patients who underwent RD repair with concomitant use of MTI therapy had a decreased risk of subsequent RD and need for further ocular intervention, including pars plana vitrectomy and complex RD repair compared with matched patients not using MTI therapy.These findings suggest that modulating mTOR and downstream signaling may present a therapeutic target for proliferative vitreoretinopathy prevention after primary RD repair; additional studies exploring the potential efficacy and safety of these agents in this setting should be considered.
