Clonal Hematopoiesis and Risk of Trastuzumab-Related Cardiotoxic Effects

What is the role of clonal hematopoiesis of indeterminate potential (CHIP) in trastuzumab-related cardiotoxic effects?In this cohort study, the UK Biobank cohort including 15 729 patients with breast cancer, trastuzumab treatment was associated with incident heart failure, the risk was amplified in individuals with CHIP, particularly in those with larger clones; in an independent cohort of 454 patients with breast cancer treated with trastuzumab at Seoul National University Hospital, CHIP was independently associated with cardiotoxic effects. In addition, -deficient clonal hematopoiesis exacerbated trastuzumab-induced left ventricular systolic dysfunction in a mouse model.These findings identified CHIP as significantly associated with trastuzumab-related cardiotoxic effects.
