Is lower posttreatment amyloid burden in TRAILBLAZER-ALZ 2 associated with Alzheimer disease (AD) clinical benefit?In this secondary analysis of a randomized clinical trial, lower posttreatment amyloid burden was correlated with slower cognitive and functional decline in participants with early symptomatic AD and with decreases in AD-related plasma biomarkers, including phosphorylated tau and glia fibrillary protein.The correlation between posttreatment amyloid level and clinical benefit observed in this trial supports amyloid plaque removal as the mechanism of action for donanemab treatment.
