Deuterated Pentoxifylline Analog in the Treatment of Necrobiosis Lipoidica

Is PCS499, an oral deuterated analog of pentoxifylline, safe and effective in improving clinical outcomes in adults with necrobiosis lipoidica (NL)?In this phase 2, open-label nonrandomized clinical trial of 12 adults with biopsy-confirmed NL, the most common adverse event was mild gastrointestinal symptoms. Among 10 participants who completed treatment, clinical improvements were observed across clinician- and patient-reported measures, including lesion activity and quality of life; the 2 participants with ulcerated lesions achieved complete closure.PCS499 may be safe and may show clinical benefit in NL, with notable responses observed in ulcerated disease, supporting further evaluation in larger, controlled studies.
