Adjunctive Antipsychotics in Major Depressive Disorder

How do US Food and Drug Administration (FDA)–approved atypical antipsychotics in major depressive disorder (MDD) compare with respect to overall efficacy and acceptability?In this systematic review and network meta-analysis, a hierarchy of efficacy and acceptability was observed, in which lumateperone had the highest effect size for efficacy, followed by aripiprazole, and in which aripiprazole had the highest overall acceptability, followed by cariprazine. Secondary and exploratory outcomes accorded with the coprimary outcomes across measures of efficacy, acceptability, and tolerability.FDA-approved atypical antipsychotic agents for MDD exhibit differential efficacy, acceptability, and tolerability relevant to treatment selection and sequencing.
