Concurrent Changes in Plasma Phosphorylated Tau 217, Tau PET, and Cognition in Preclinical Alzheimer Disease

Are plasma phosphorylated tau 217 (p-tau217) and tau positron emission tomography (PET) suitable biomarkers to detect disease modification in preclinical Alzheimer disease clinical trials?In this diagnostic/prognostic study, changes in tau PET uptake in frontoparietal regions were strongly associated with concurrent changes in cognition. Changes in plasma p-tau217 were more modestly associated with concurrent cognitive changes.These findings highlight the value of tau PET for real-time tracking of disease progression to enhance detection of disease-modifying effects in preclinical Alzheimer disease trials.
