Vitamin D Receptor Polymorphisms and the Effect of Vitamin D Supplementation on Diabetes Risk Among Adults With Prediabetes

How do polymorphisms in the vitamin D receptor influence the effect of vitamin D supplementation on diabetes risk among adults with prediabetes?In this genetic association study using data from the D2d study, 618 adults with the AA genotype of the ApaI polymorphism (rs797523) did not respond to treatment with 4000 IU/d of vitamin D, compared with placebo (hazard ratio [HR], 1.02 [95% CI, 0.72-1.44]). In contrast, 1480 adults with AC and CC genotypes had a reduced risk of diabetes (HR, 0.81 [95% CI, 0.66-0.99]).This study suggests that ApaI genotyping may identify adults with prediabetes who experience benefits from high-dose vitamin D supplementation to lower their risk of developing diabetes.
