Antibody-Drug Conjugates for Locally Advanced and Metastatic Urothelial Carcinoma

How do clinical outcomes associated with antibody-drug conjugates for locally advanced or metastatic urothelial carcinoma compare across clinical settings?In this systematic review and network meta-analysis of 40 studies, enfortumab vedotin monotherapy was associated with consistent objective response rates across interventional (43.9%) and observational (44.6%) cohorts. Enfortumab vedotin plus pembrolizumab was associated with high response rates in total (67.5%) and cisplatin-ineligible (65.4%) populations, as was disitamab vedotin plus immunotherapy across interventional (74.7%) and observational (61.7%) cohorts.These findings suggest that enfortumab vedotin–based regimens are associated with robust outcomes across clinical settings, and disitamab vedotin combinations are associated with potent clinical activity.
