Orforglipron Added to Titrated Insulin Glargine in Type 2 Diabetes

What is the efficacy and safety of once-daily oral orforglipron vs placebo when added to titrated insulin glargine in adults with inadequately controlled type 2 diabetes?In this randomized clinical trial including 546 participants, the mean change in hemoglobin A at 40 weeks was −1.58% with orforglipron, 3 mg once daily, −1.88% with orforglipron, 12 mg once daily, −1.82% with orforglipron, 36 mg once daily, and −0.79% with placebo. Each dosage of orforglipron was superior to placebo and did not increase hypoglycemia risk.Orforglipron improved glycemic control compared with placebo when added to titrated basal insulin in adults with type 2 diabetes.
