Ultralow-Dose Interleukin 10–Expressing Chimeric Antigen Receptor T Cells in Relapsed/Refractory Diffuse Large B-Cell Lymphoma

What is the safety and efficacy of interleukin 10–expressing CD19 chimeric antigen receptor (CAR) T cells (META 10-19) in adults with relapsed/refractory (R/R) diffuse large B-cell lymphoma (DLBCL)? In this phase 1, nonrandomized clinical trial involving 13 patients with R/R DLBCL, ultralow-dose META 10-19 (2 × 10 to 2 × 10 CAR T cells/kg) achieved an objective response rate of 92.3% and complete remission rate of 84.6% among all patients who received a META 10-19 infusion, with cytokine release syndrome and immune effector cell–associated neurotoxicity syndrome being predominantly grade 1 or 2. The results of this nonrandomized clinical trial suggest that ultralow-dose META 10-19 demonstrated promising antitumor activity, with manageable safety profiles in patients with R/R DLBCL.
